Enola (Milly Bobby Brown) ist die Nachzüglerin der Familie Holmes. Ihr Vater verstarb, als sie noch sehr klein war, ihre großen Brüder Mycroft (Sam Claflin) und Sherlock (Henry Cavill) sind kurz darauf zuhause ausgezogen; Enola ist allein mit ihrer Mutter Eurdoria (Helena Bonham-Carter) aufgewachsen. Es gab in ihrem ganzen Leben daher bisher nur dieses Haus, ihre Mutter und ein Hausmädchen. Dadurch ist Enola unbeeinflusst von den gesellschaftlichen Erwartungen im viktorianischen England herangewachsen und hat außerdem eine außergewöhnliche Erziehung genossen: Jedes Buch in der Bibliothek hat sie gelesen, sie studierte vor allem Geschichte und Naturwissenschaften, lernte Kampfsportarten. Aber sie hat auch kaum eine Ahnung, wie es in der wirklichen Welt zugeht. An ihrem 16. Geburtstag verschwindet ihre Mutter nun spurlos und ihre Brüder wollen sie daraufhin in ein Internat stecken. Aber nicht mit Enola! Ihre Mutter hat ihr beigebracht, dass sie auf eigenen Füßen stehen muss und deshalb macht sie sich auf den Weg nach London, um ihre Mutter zu suchen.
